为探讨高糖和脂多糖(LPS)刺激下人脐静脉内皮细胞(HUVEC)TLR4的表达及高糖促进动脉粥样硬化与TLR4的关系,我们用RT-PCR和流式细胞术分别测定了高糖和LPS刺激下HUVECTLR4 mRNA和蛋白的表达;用ELISA测定高糖和LPS刺激下HUVECMCP-1的产生。研究结果表明,高糖上调HUVECTLR4 mRNA及其蛋白的表达,促进HU-VEC分泌MCP-1;高糖协同LPS上调HUVEC细胞TLR4蛋白的表达,使HUVEC分泌MCP-1进一步增加。研究发现,高糖促进动脉粥样硬化与TLR4有关;高糖可能协同感染因素促进动脉粥样硬化的发生。
To investigate the expression of TLR4 in human umbilical vein endothelial cells (HUVECs) stimulated with high glucose and lipopolysaccharide (LPS) and the relationship between high glucose-induced atherosclerosis and TLR4, RT-PCR and flow cytometry The expression of HUVECTLR4 mRNA and protein under the stimulation of sugar and LPS; the production of HUVECMCP-1 under high glucose and LPS stimulation was measured by ELISA. The results showed that high glucose up-regulated the expression of HUVECTLR4 mRNA and protein, and promoted the secretion of MCP-1 by HU-VEC. High glucose and LPS up-regulated the expression of TLR4 protein in HUVEC and further increased the secretion of MCP-1 by HUVEC. Study found that high glucose promote atherosclerosis and TLR4 related; high glucose may synergize with infection factors to promote the occurrence of atherosclerosis.
