目的:探讨microRNA143(miR-143)在膀胱移行细胞癌中的表达及其对膀胱癌细胞系生物学行为的影响。方法:实时定量RT-PCR法验证miR-143在膀胱癌中的表达。应用化学方法合成成熟miR-143(miR-143mimics),瞬时转染J82、T24细胞,分别通过流式细胞计数和MTS检测miR-143对J82、T24细胞凋亡和增殖的影响。结果:miR-143在膀胱肿瘤中普遍低表达,体外转染miR-143能抑制J82、T24细胞生长,但对细胞凋亡无影响。结论:MiR-143可能参与膀胱癌的发生发展。
Objective: To investigate the expression of microRNA143 (miR-143) in bladder transitional cell carcinoma and its effect on the biological behavior of bladder cancer cell lines. Methods: Real-time quantitative RT-PCR was used to confirm the expression of miR-143 in bladder cancer. The mature miR-143 (miR-143mimics) was synthesized and transiently transfected into J82 and T24 cells. The apoptosis and proliferation of J82 and T24 cells were detected by flow cytometry and MTS respectively. Results: miR-143 was overexpressed in bladder tumor. MiR-143 transfected in vitro inhibited the growth of J82 and T24 cells but had no effect on apoptosis. Conclusion: MiR-143 may be involved in the development of bladder cancer.
