目的 探讨单侧视觉剥夺后鸽子两侧视盖基因的差异表达情况。方法 建立单眼剥夺动物模型 ,抽提、纯化左右视盖mRNA ,利用SSH方法建立左侧视盖差异表达文库 ,对其中EST进行克隆、测序 ,反Northern杂交去除假阳性。结果 共筛选得到 12个差异表达片段 ,经同源性检索 ,其中 11个未发现有高同源性的基因 ,提示可能为新的EST ;另一差异片段与CGI- 3 1蛋白有 85 %的同源性。结论 视觉相关脑区差异表达序列的分析可能为视觉形成的分子机制或视觉通路的分子构筑提供有益的线索
Objective To investigate the differential expression of the gene on both sides of pigeons after unilateral visual deprivation. Methods The animal model of monocular deprivation was established. The left and right visual cortex mRNAs were extracted and purified. The left visual cortex differential expression library was established by SSH method. The ESTs were cloned, sequenced and reverse Northern blot hybridization was used to remove false positives. Results A total of 12 differentially expressed fragments were screened by homology, 11 of which were not found to have high homology, suggesting that they might be new ESTs. Another difference fragment was 85% homologous to CGI-3 protein Sex. Conclusions The analysis of differentially expressed sequences in the brain regions of visual relevance may provide useful clues to the molecular mechanism of visual formation or the molecular construction of visual pathways
