孕激素在妊娠的建立和维持过程中起着非常重要的作用,但目前为止未见有关大鼠子宫孕激素受体(PR)基因在该过程中表达情况的系统报道。本文用反转录偶联聚合酶链式反应(RT-PCR)方法测定了大鼠动情周期和妊娠早期(d1~d10)子宫孕激素受体基因的转录,结果表明:1.动情周期中,子宫PRmRNA水平在动情期最高,在动情后期最低,动情期水平约为动情后期的2倍。2.妊娠开始后,子宫PRmRNA水平迅速上升,于d3~d4(着床前)达到高峰,约为d1的2倍,而于d6(着床中)明显下降至d1的50%左右,然后维持此低水平至d10。3.孕后d7~d10,子宫着床部位的PRmRNA水平明显高于非着床部位。我们对这些变化的生理意义作了初步讨论。
Progesterone plays a very important role in the establishment and maintenance of pregnancy, but so far no systematic report on the expression of rat progesterone receptor (PR) gene in the process has been reported. In this study, the transcription of uterine progesterone receptor gene in estrous cycle and early gestation (d1 ~ d10) was determined by reverse transcription-polymerase chain reaction (RT-PCR). The results showed that: 1. During the estrous cycle, the level of uterine PR mRNA was the highest in estrus and lowest in late estrus. The level of estrus was about twice as high as that in late estrus. 2. After the start of pregnancy, uterine PR mRNA levels increased rapidly and reached a peak at d3 ~ d4 (pre-implantation), which was about double that of d1, but dropped to about 50% of d1 at d6 (implantation), and then maintained at a low level Level to d10.3. D7 ~ d10 after pregnancy, uterine implantation site PR mRNA levels were significantly higher than non-implantation site. We made a preliminary discussion of the physiological significance of these changes.
