对绿菜花“Galaxy”、“bc-0”和“bc-4”3个基因型进行了渗透压、外源激素、热激处理温度和时间、花蕾长度或小孢子发育时期的影响因素研究。结果表明,激素的作用并不明显,但相比之下,单独加入6-BA比同时加入6-BA、NAA效果好;绿菜花游离小孢子培养的最适热激处理温度和时间为32.5℃,24h;绿菜花游离小孢子的最适培养花蕾长度因基因型而异,如基因型“bc-4”蕾长为4.1~5.0mm,此时大多数小孢子处在单核晚期、双核早期,出胚率高达37.08胚/蕾
The genotypes of “Galaxy”, “bc-0” and “bc-4” were studied on the factors of osmotic pressure, exogenous hormones, temperature and time of heat shock treatment, flower bud length or microspore development. The results showed that the role of hormones was not obvious, but compared with 6-BA alone, 6-BA and NAA were better. The optimum heat-treatment temperature and time of green microspore culture were 32. 5 ℃, 24h. The optimal length of flower buds of green microspores was different according to genotypes. For example, the length of genotype “bc-4” was 4.1-5.0mm, at which point most microspores were in single Early nuclear, early dual nucleus, the embryo rate as high as 37.08 embryos / bud
