为了解血小板来源生长因子(PDGF)与IgA肾病(IgAN)的临床及病理改变之间的关系,我们检测了10例IgAN和2例健康对照肾小球中PDGF mRNA和蛋白质、PDGFβ受体的表达。 研究对象 10例IgAN均为肾活检确诊的原发性IgAN,男、女各5例,年龄13~53岁。2例对照为肾移植供肾标本,经光镜组化排除肾病变。 方法 取肾活检组织,微分离出肾小球后以原位反转录PCR法测肾小球中PDGFβ链mRNA。PDGF和PDGFβ受体的蛋白质均用四层PAP法检测。 结果 IgAN的临床、病理资料和PDGF的检测结
To investigate the relationship between platelet-derived growth factor (PDGF) and the clinical and pathological changes of IgA nephropathy (IgAN), we examined the expression of PDGF mRNA and protein and PDGFβ receptor in glomerular of 10 IgAN and 2 healthy controls . All 10 IgAN cases were primary IgAN diagnosed by renal biopsy, including 5 males and 5 females, aged 13-53 years. Two cases of kidney transplantation for kidney transplantation for renal specimens, histological exclusion of renal lesions. Methods Kidney biopsy was performed to detect the PDGFβ chain mRNA in the glomerulus by microinjection of glomerulus by in situ reverse transcription PCR. The PDGF and PDGFβ receptor proteins were detected by the four-layer PAP assay. Results IgAN clinical, pathological data and PDGF test results
