目的观察猴免疫缺陷病毒(SIV)感染CEM×174细胞后CD28家族免疫调控分子CD28、BTLA、ICOS、PD-1 mRNA表达水平的动态变化。方法以SIVmac251感染接种CEM×174细胞,收集感染后12,24,48,72,96,120,144,168,216h共8个时间点及正常未感染细胞,提取细胞RNA,以染料法逆转录荧光定量PCR法检测SIV、CD28、ICOS、PD-1、BTLA mRNA的表达。结果SIV mRNA在48h时表达即显著升高(P<0.05),随着SIV mRNA量的逐渐上升,ICOS在48h时开始显著升高(P<0.05),而CD28、PD-1 mRNA在120h时才出现表达显著增加(P<0.05);BTLA mRNA则呈现下降趋势,并在168h开始出现显著下调(P<0.05)。结论除BTLA以外,所检测SIV感染后细胞中促进免疫激活和抑制免疫的免疫调节因子(ICOS、PD-1、CD28)表达均显著升高,提示AIDS中存在免疫紊乱。
Objective To observe the dynamic changes of CD28, BTLA, ICOS and PD-1 mRNA expressions of CD28 family members after infection with simian immunodeficiency virus (SIV) in CEM × 174 cells. Methods SIVmac251 cells were inoculated with CEM × 174 cells, and 8, 12, 24, 48, 72, 96, 120, 144, 168 and 216 h post infection were collected for 8 time points and normal uninfected cells. RNA was extracted and SIV was detected by reverse transcription polymerase chain reaction (RT- CD28, ICOS, PD-1, BTLA mRNA expression. Results The expression of SIV mRNA was significantly increased at 48h (P <0.05). With the increase of SIV mRNA level, ICOS began to increase significantly at 48h (P <0.05), while the expression of CD28 and PD-1 mRNA at 120h (P <0.05). The expression of BTLA mRNA showed a downward trend and began to show a significant decrease at 168h (P <0.05). Conclusion The expression of ICOS, PD-1 and CD28, which promote immune activation and immune suppression, were significantly increased in cells except for BTLA, suggesting that immune disorders exist in AIDS.
