目的观察姜黄素诱导HO-1高表达及其对动脉粥样硬化(AS)大鼠血清中氧化低密度脂蛋白(ox-LDL)、丙二醛(MDA)含量的影响。方法将34只健康♂Wistar大鼠,随机分为正常对照组(n=8)、模型组(n=10)、姜黄素组(n=8)及抑制组(n=8),模型组、姜黄素组及抑制组同法复制AS模型,姜黄素组加用姜黄素,抑制组加用姜黄素及锌原卟啉Ⅸ;第14周末处死所有大鼠,取降主动脉检测HO-1的表达、分布及活力,经心脏取血检测ox-LDL、MDA的含量。结果姜黄素组较模型组的HO-1表达量及活力明显增强(P<0.05),ox-LDL、MDA含量显著降低(P<0.05);抑制组较姜黄素组的HO-1表达量及活力明显下降(P<0.05),ox-LDL、MDA含量显著增高(P<0.05)。结论姜黄素可诱导HO-1高表达并显著减轻脂质过氧化程度,锌原卟啉Ⅸ抑制HO-1高表达可使姜黄素抗脂质过氧化作用显著下降。
Objective To observe the high expression of HO-1 induced by curcumin and its effect on serum levels of ox-LDL and malondialdehyde (MDA) in rats with atherosclerosis (AS). Methods Forty healthy Wistar rats were randomly divided into normal control group (n = 8), model group (n = 10), curcumin group (n = 8) and inhibition group (n = 8) Curcumin group and curcumin group, curcumin group and zinc protoporphyrin Ⅸ; At the end of the 14th week, all the rats were sacrificed, and the descending aorta was taken to detect HO-1 Expression, distribution and vitality, the content of ox-LDL and MDA in the heart blood was measured. Results Compared with the model group, the expression of HO-1 in curcumin group was significantly increased (P <0.05) and the levels of ox-LDL and MDA in the curcumin group were significantly decreased (P <0.05). Compared with the curcumin group, the expression of HO- (P <0.05), and the levels of ox-LDL and MDA were significantly increased (P <0.05). Conclusion Curcumin can induce high expression of HO-1 and significantly reduce the degree of lipid peroxidation. Inhibition of lipid peroxidation by curcumin by Zn protoporphyrin IX inhibits the expression of HO-1 significantly.
