本文通过聚苯乙烯固载聚乙二醇氨基树脂与对羟甲基苯氧乙酸结合制备了具有酸敏手臂结构的对羟甲基苯氧乙酰胺PEG树脂(简称PS-PEG-HMPA)。在合成中改进了PS-PEG-NH2树脂的制备方法,提高了树脂上功能基的转化率,不必进行封闭剩余活性基的操作,就得到具有单一功能基的树脂。以此树脂为载体,运用Fmoc保护化学固相肽合成方法,合成了白蛋白八肽片段KQTALYYC。纯化后的肽用HPLC进行纯度分析。氨基酸组成分析的实测值也与理论值相符合,确证了产物的均一性。
In this paper, PS-PEG-HMPA (PEG-HMPA) with acid-sensitive arm structure was prepared by the combination of polystyrene-immobilized polyethylene glycol amino resin and p-hydroxymethylphenoxyacetic acid. In the synthesis, the preparation method of PS-PEG-NH2 resin is improved, the conversion rate of the functional groups on the resin is improved, and the resin with a single functional group is obtained without the operation of blocking the remaining active groups. Using this resin as a carrier, the albumin octapeptide fragment KQTALYYC was synthesized by Fmoc-protected chemical solid-phase peptide synthesis. Purified peptides were subjected to purity analysis by HPLC. The amino acid composition analysis of the measured value is also consistent with the theoretical value, confirming the product uniformity.
