目的研究实验性脾虚证大鼠胰岛细胞胰岛素、胰岛淀粉样多肽、胰高血糖素和生长抑素分泌活动的变化,探讨中医学脾虚证发病与胰岛激素分泌活动的关系。方法用Wistar雄性成年大鼠24只,分为正常对照组、实验性脾虚组、自然恢复组和中药治疗组。4组动物同时麻醉处死,取胰尾固定于Bouin液,制成石蜡切片。用免疫组织化学法显示胰岛B细胞胰岛素和胰岛淀粉样多肽、A细胞胰高血糖素和D细胞生长抑素,并用显微分光光度计对免疫染色强度进行了定量测定。结果与对照组相比,脾虚组胰岛B细胞胰岛素和A细胞胰高血糖素免疫染色明显减弱,而D细胞生长抑素和B细胞胰岛淀粉样多肽免疫染色明显增强。结论上述结果提示,胰岛细胞激素分泌活动失常可能与脾虚证代谢异常有密切关系。
Objective To study the changes of insulin, amylin, glucagon and somatostatin secretion in islet cells of rats with experimental spleen deficiency syndrome and to explore the relationship between the pathogenesis of insulin deficiency and spleen deficiency syndrome in traditional Chinese medicine. Methods Twenty-four male Wistar rats were divided into normal control group, experimental spleen deficiency group, natural recovery group and traditional Chinese medicine treatment group. Four animals were anesthetized and sacrificed at the same time, and the pancreatic tail was fixed in Bouin liquid to make paraffin sections. Immunohistochemistry was used to detect islet B cell insulin and amylin, A-cell glucagon and D-cell somatostatin, and the intensity of immunostaining was quantitatively determined with a spectrophotometer. Results Compared with the control group, insulin and A cell glucagon immunostaining of pancreatic islet B cells in spleen deficiency group was significantly weakened, whereas the somatostatin D cells and islet amyloid peptide immunostaining of B cells were significantly increased. Conclusions The above results suggest that abnormalities of islet cytokine secretion may be closely related to the abnormal metabolism of spleen deficiency syndrome.
