目的 探讨 0 75mM丁酸钠对肝卵圆细胞甲胎蛋白 (AFP)表达和向细胞外分泌AFP的影响。方法从喂饲含 0 1%乙硫氨酸的胆碱缺乏性饮食 4~ 6周的大鼠肝脏中分离出肝卵圆细胞。噻唑蓝 (MTT)比色法做生长曲线 ,观察 0 75mM丁酸钠对肝卵圆细胞生长的影响。收集 0 75mM丁酸钠处理 4天后的肝卵圆细胞及其培养上清液。将细胞裂解后 ,用WesternBlot检测胞内AFP的表达水平。用酶联免疫吸附法 (ELISA)测定培养上清液中的AFP浓度。结果 生长曲线的结果显示 0 75mM丁酸钠对肝卵圆细胞的生长有一定的抑制作用。WesternBlot的结果提示0 75mM丁酸钠能显著降低肝卵圆细胞AFP的表达水平 (P <0 0 5 ) ,但ELISA法测定的培养上清液中AFP的含量却没有显著改变。结论 0 75mM丁酸钠对肝卵圆细胞的生长有一定的抑制作用 ,并能降低肝卵圆细胞AFP的表达水平。
Objective To investigate the effect of 0 75 mM sodium butyrate on the expression of AFP and the secretion of AFP from hepatic oval cells. Methods Hepatic oval cells were isolated from rat livers fed a choline-deficient diet containing 0.1% ethionine for 4 to 6 weeks. MTT assay was used to observe the growth of hepatic oval cells at 0 75 mM sodium butyrate. Hepatic oval cells and their culture supernatants were collected after 4 days of 075 mM sodium butyrate treatment. After the cells were lysed, the expression level of intracellular AFP was detected by Western Blotting. AFP concentrations in culture supernatants were determined by enzyme-linked immunosorbent assay (ELISA). RESULTS The results of the growth curve showed that 0 75 mM sodium butyrate had a certain inhibitory effect on the growth of hepatic oval cells. Western Blot results suggest that 75 mM sodium butyrate can significantly reduce the level of AFP expression in hepatic oval cells (P <0.05), but the content of AFP in the culture supernatant assayed by ELISA did not change significantly. Conclusions 0 75 mM sodium butyrate can inhibit the growth of hepatic oval cells and reduce the expression of AFP in hepatic oval cells.
