[目的]探讨不同血液乙醇浓度对家兔脑基底动脉内皮细胞热休克蛋白70(HSP70)表达的影响。[方法]通过灌胃的方法建立血液乙醇浓度梯度的家兔动物模型,用气相色谱仪测定各灌胃量组血液乙醇浓度;通过免疫组化的方法检测脑基底动脉内皮细胞HSP70的表达。[结果]家兔灌酒后血液乙醇浓度持续增高,灌胃后1.5~2h,血液乙醇浓度达到峰值;灌酒后血液乙醇浓度与灌酒量存在正相关关系。各酒精灌胃组脑基底动脉内皮细胞HSP70均有表达,且表达量随灌酒量的增加而增多,各组之间差异存在统计学意义(P﹤0.05)。[结论]家兔脑基底动脉内皮细胞的应激反应程度与血液乙醇浓度存在正相关关系(r=0.969,P﹤0.05)。
[Objective] To investigate the effect of different blood alcohol concentrations on the expression of heat shock protein 70 (HSP70) in basilar artery endothelial cells of rabbits. [Method] Rabbit animal model of blood ethanol concentration gradient was established by intragastric administration. The blood ethanol concentration in each gavage group was determined by gas chromatography. The expression of HSP70 in basilar artery endothelial cells was detected by immunohistochemistry. [Result] The blood ethanol concentration in rabbits increased continuously. After 1.5 to 2 hours after gavage, the blood ethanol concentration peaked. There was a positive correlation between blood alcohol concentration and alcohol consumption after filling. The expressions of HSP70 in basal artery endothelial cells of various alcoholic gavages groups were all increased, and the expression of HSP70 increased with the increase of alcohol consumption. The differences among the groups were statistically significant (P <0.05). [Conclusion] The degree of stress response in basilar artery endothelial cells of rabbits has a positive correlation with the blood alcohol concentration (r = 0.969, P <0.05).
