目的:观察酸味中药五味子对来亨鸡维生素A缺乏型干眼症的疗效及眼角膜组织中维生素A结合蛋白(RBP)、维生素A结合蛋白受体(STRA6)的表达变化,探讨酸味中药缓解干眼症的作用机制。方法:将180只白色来亨鸡随机分为正常组、模型组(维生素A缺乏组)、维生素A对照组、五味子治疗组(高、中、低剂量组)。分别于用药前、用药后2周、用药后4周对各组来亨鸡进行Schirmer I试验(SIT)和泪膜破裂时间(BUT)检查。采用免疫印迹法检测五味子对眼角膜组织的维生素A结合蛋白(RBP)及维生素A结合蛋白受体(STRA6)蛋白表达的影响。结果:用药2周、4周后,与模型组相比,五味子各剂量组Schirmer试验滤纸湿长有明显增加,泪膜破裂时间逐渐增加。与正常组比较,模型组RBP、STRA6蛋白表达显著降低,五味子各剂量组作用4周后,显著上调RBP、STRA6蛋白表达。结论:五味子可以提高泪液基础分泌量,维持泪膜稳定性,并促进RBP、STRA6蛋白表达,从而减轻干眼症状。
OBJECTIVE: To observe the curative effect of sour herb Schisandra chinensis on vitamin A-deficiency dry eye syndrome and the expression of retinal ganglion (RBP) and vitamin A-binding protein receptor (STRA6) Symptoms of the mechanism of action. Methods: 180 white Leghorn chickens were randomly divided into normal group, model group (vitamin A deficiency group), vitamin A control group and Schisandra chinensis treatment group (high, medium and low dose group). The Schirmer I test (SIT) and tear film break-up time (BUT) were performed in each group before treatment, 2 weeks after treatment and 4 weeks after treatment. Western blotting was used to detect the effects of Schisandra chinensis on the protein expression of retinal ganglion (RBP) and retinoicin A receptor (STRA6) in corneal tissue. Results: Compared with the model group, the Schirmer test filter paper’s wet length significantly increased and the tear film’s rupture time gradually increased after 2 and 4 weeks of treatment. Compared with the normal group, the expression of RBP and STRA6 protein in the model group decreased significantly, and the expression of RBP and STRA6 protein increased significantly in each dose group of Schisandrae for 4 weeks. Conclusion: Schisandra can improve the basic tear secretion, maintain tear film stability, and promote RBP, STRA6 protein expression, thereby reducing dry eye symptoms.
