目的:研究金耳多糖(TMP)对豚鼠哮喘模型支气管平滑肌收缩功能的影响,初步分析其作用机制。方法:采用肺阻力(RL)和肺动态顺应性(Cdyn)指标评价TMP对致敏豚鼠抗原攻击前后肺功能变化的影响;采用离体气管条法观察TMP对豚鼠气道的直接扩张和对抗氨甲酰胆碱(CCH)和组胺引起的气道平滑肌收缩作用。结果:在整体试验中,TMP 1.60g/kg预处理,能明显抑制致敏豚鼠抗原攻击后引起的RL增加和Cdyn下降;在离体试验中,5-80mg/mL TMP对豚鼠气道具有直接舒张作用,10-80mg/mL TMP对CCH引起的气道平滑肌收缩呈现非常显著的抑制作用(P<0.05),但对组胺引起的气道平滑肌收缩无对抗作用。结论:TMP抑制豚鼠哮喘模型肺功能下降可能与CCH作用有关。
Objective: To study the effects of polysaccharides from Ginkgo biloba (TMP) on the contractile function of bronchial smooth muscle in guinea pig asthma model and to analyze its mechanism. Methods: The pulmonary function (RL) and dynamic lung compliance (Cdyn) index were used to evaluate the effect of TMP on pulmonary function before and after challenge with antigen. The direct expansion of TMP on guinea pig airway Chlamydia (CCH) and histamine-induced airway smooth muscle contraction. Results: In the whole experiment, TMP 1.60g / kg pretreatment significantly inhibited the increase of RL and Cdyn induced by antigen challenge in guinea pigs; in vitro, 5-80mg / mL TMP had a direct effect on the airway of guinea pigs The diastolic effect, 10-80mg / mL TMP had a very significant inhibitory effect on airway smooth muscle contraction induced by CCH (P <0.05), but had no antagonistic effect on histamine-induced airway smooth muscle contraction. Conclusion: TMP may reduce the lung function of guinea pig asthma model may be related to the role of CCH.
