目的研究左归降糖舒心方对转基因2型糖尿病MKR鼠心肌损伤的保护作用。方法以MKR鼠为研究对象,ig左归降糖舒心方对其进行30 d治疗,设野生C57BL/6鼠同期对照。用透射电镜观察小鼠心肌形态学改变;比色法测定心肌中丙二醛(MDA)的量和超氧化物歧化酶(SOD)的活性;全自动生化分析仪检测心功能指标肌酸激酶同工酶(CKMB)和乳酸脱氢酶(LDH)活性。结果经左归降糖舒心方治疗后,MKR鼠心肌组织形态学病变减轻,心肌细胞线粒体嵴部分消失,肌纤维排列较整齐;心肌组织MDA量下降(P<0.05、0.01),SOD活性升高(P<0.01),且呈剂量依赖性;血清CKMB和LDH活性下降(P<0.05、0.01),且呈剂量依赖性。结论左归降糖舒心方可通过抗脂质过氧化,对MKR转基因2型糖尿病MKR鼠心肌产生保护作用。
Objective To investigate the protective effect of Zuo Gui Jiang Tang Shu Xin Fang on myocardium injury in transgenic MKR mice. Methods MKR mice were used as study subjects. ig Zuo Gui Jiang Tang Shu Xin Fang was treated for 30 days. The wild C57BL/6 mice were used for the same period of time. The morphological changes of the myocardium were observed by transmission electron microscopy; the content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in the myocardium were determined by colorimetric method; the cardiac function markers of creatine kinase were measured by automatic biochemical analyzer. Chemical enzyme (CKMB) and lactate dehydrogenase (LDH) activity. Results After treated with Zuogui Jiangtang Shuxin Decoction, the morphological lesions of myocardium in MKR mice were alleviated, the myocardial mitochondria were partially disappeared, the muscle fibers were arranged neatly, the myocardial tissue MDA content decreased (P<0.05, 0.01), and the activity of SOD increased ( P <0.01), and in a dose-dependent manner; serum CKMB and LDH activity decreased (P <0.05, 0.01), and in a dose-dependent manner. Conclusion Zuo Gui Jiang Tang Shu Xin Fang can protect the myocardium of MKR transgenic MKR mice by anti-lipid peroxidation.
