目的 探索桥本甲状腺炎与Graves病患者甲状腺滤泡上皮细胞异常表达的HLAⅡ类分子能否递呈自身抗原。方法 标记链霉亲和素结合 (LSAB)技术检测患者甲状腺滤泡上皮细胞中HLAⅡ类分子、不变链 (Ii链 )与B7分子的表达。原位杂交技术 (ISH)进一步从mRNA水平上检测Ii链的表达。结果 桥本甲状腺炎与Graves病患者甲状腺滤泡上皮中HLAⅡ类分子、Ii链与B7分子表达的格局呈高度异质性。结论 至少有一部分自身免疫性甲状腺炎患者的滤泡上皮细胞具备递呈自身抗原必要的分子基础。
Objective To explore if Hashimoto’s thyroiditis and thyroid follicular epithelial cells in patients with Graves disease abnormal expression of HLA class II molecules can present their own antigens. Methods The expression of HLA class II molecules, I chain and B7 in thyroid follicular epithelial cells was detected by streptavidin - binding assay (LSAB). In situ hybridization (ISH) further detects mRNA expression of the I chain. Results Hashimoto’s thyroiditis and Graves disease thyroid follicular epithelial HLA class II molecules, Ii chain and B7 expression pattern was highly heterogeneous. Conclusions At least some of the follicular epithelial cells in patients with autoimmune thyroiditis have the necessary molecular basis to present autoantigens.
