[目的]研究HPV16 E6和Hippo途径效应分子YAP的相关性。[方法]用Western Blot、RT-PCR和激光共聚焦显微镜观察HPV16 E6过表达或敲减后YAP的表达和定位,以及其直接作用的下游基因结缔组织生长因子(CTGF)的表达。[结果]HPV16 E6敲减或过表达时YAP相应地表达减少或增加,磷酸化的YAP增加或减少,YAP主要定位于胞浆或胞浆和胞核中,YAP下游CTGF的表达相对应减少或增加。[结论]HPV16 E6诱导Hippo途径失常,YAP的表达升高,可能提升HPV16 E6的致癌性,Hippo途径可能是治疗高危性HPV感染相关疾病的候选药物作用靶标。
[Objective] To study the correlation between HPV16 E6 and Hippo pathway effector molecule YAP. [Method] The expression and localization of YAP after HPV16 E6 overexpression or knockdown were detected by Western Blot, RT-PCR and confocal laser scanning microscope, and the direct expression of connexin tissue growth factor (CTGF). [Result] The expression of YAP was decreased or increased correspondingly when HPV16 E6 knocked down or overexpressed, the phosphorylated YAP increased or decreased, YAP mainly located in the cytoplasm, cytoplasm and nucleus, and the expression of CTGF in the downstream of YAP decreased or increase. [Conclusion] The HPV16 E6 induces the abnormal Hippo pathway and the increased expression of YAP, which may increase the carcinogenicity of HPV16 E6. Hippo pathway may be the target of drug candidates for the treatment of diseases related to high risk HPV infection.
