目的:探讨甘露消毒丹对柯萨奇病毒A16型(CoxA16)小鼠模型的影响。方法:7日龄ICR小鼠150只,随机分为正常组、模型组、阿昔洛韦组、甘露消毒丹组,每组25只;除正常组外,其余各组每只小鼠腹腔内注射20ul 107TCID50的CoxA16标准株原液进行造模,造模后的第5天,除正常组和模型组外,其余4组使用相应的药物干预治疗。观察小鼠临床表现;在光镜下观察肌肉组织形态学变化;用细胞增殖与毒性检测试剂盒(CCK-8)检测肌肉匀浆液对Hela细胞存活率的变化;电镜观察肌肉组织病毒颗粒的变化,采用ELISA法检测肌肉组织IL-6、IL-10、INF-γ的浓度。结果:与模型组比较,发现阿昔洛韦及甘露消毒丹(24.4g/kg、12.2g/kg、6.1g/kg)均可提高小鼠的生存率并使体重恢复到正常情况。HE染色和电镜结构表明甘露消毒丹(24.4g/kg、12.2g/kg、6.1g/kg)和阿昔洛韦均可减轻CoxA16病毒对肌肉的损伤,并减少肌肉内病毒的颗粒。通过肌肉组织匀浆液对Hela细胞存活率的变化表明甘露消毒丹能抑制病毒在肌肉内的生长。ELISA检测结果发现阿昔洛韦与甘露消毒丹均能减少IL-6的浓度,并提高IL-10、INF-γ的表达。结论:甘露消毒丹对CoxA16感染的小鼠有保护作用,能够抑制并杀死体内的CoxA16病毒。
Objective: To investigate the effect of Ganlu Disinfectant on CoxA16 mouse model. Methods: 150 ICR mice of 7 days old were randomly divided into normal group, model group, acyclovir group, and Ganlu Disinfectant group, with 25 rats in each group. Except normal group, Injection of 20ul 107TCID50 CoxA16 standard stock solution for modeling, the first 5 days after modeling, in addition to the normal group and model group, the remaining 4 groups using the corresponding drug intervention. The morphological changes of muscle tissue were observed under light microscope. The changes of the viability of Hela cells in muscle homogenate were detected by CCK-8 kit. The changes of the virus particles in muscle tissue were observed by electron microscope. The concentrations of IL-6, IL-10 and INF-γ in muscle tissue were detected by ELISA. Results: Compared with the model group, acyclovir and nerolidol (24.4g / kg, 12.2g / kg, 6.1g / kg) were found to improve the survival rate of mice and restore the body weight to normal. The results of HE staining and electron microscopy showed that Ganoderma lucidum (24.4g / kg, 12.2g / kg, 6.1g / kg) and acyclovir all reduced the muscle damage of CoxA16 virus and reduced the intramuscular virus particles. The change of the viability of Hela cells by the muscle tissue homogenate shows that Ganluodan can inhibit the growth of the virus in the muscle. ELISA test results showed that both acyclovir and nerolidin can reduce the concentration of IL-6, and increase the expression of IL-10, INF-γ. Conclusion: Ganlu Disinfectant can protect CoxA16-infected mice and inhibit and kill the CoxA16 virus in vivo.
