目的:研究5种人腺癌中Oct4的表达情况,为进一步探索其在肿瘤发生中的机制提供基础。方法:应用乳腺癌、胃癌、胰腺癌、肠癌、子宫内膜癌5种腺癌的组织芯片,通过免疫组织化学染色研究Oct4蛋白在组织中的表达情况,并与癌旁正常组织进行比较。结果:Oct4在5种腺癌肿瘤组织内均有表达,胃癌组织内Oct4的表达高于癌旁正常组织(P<0.05),其他腺癌组织中Oct4的表达与癌旁组织差异无统计学意义;在乳腺癌和肠癌中,无淋巴结转移患者的肿瘤组织中Oct4表达高于淋巴结转移患者的肿瘤组织(P<0.05),而在胃癌、胰腺癌及子宫内膜癌患者中未观察到这种差异;5种腺癌的肿瘤组织内,未发现Oct4的表达与临床分期有关。Oct4在癌旁正常组织中(子宫内膜癌除外)主要定位于细胞浆,而在腺癌组织中既表达于细胞核,也表达于细胞浆。结论:Oct4的表达可能是肿瘤发生过程中的早期事件,不同腺癌组织中Oct4的表达情况不完全相同,Oct4的核/浆表达模式可能与其在肿瘤发生中的作用有关。
OBJECTIVE: To study the expression of Oct4 in five human adenocarcinomas and to provide a basis for further exploration of its mechanism in tumorigenesis. Methods: The expression of Oct4 protein in tissues of breast cancer, gastric cancer, pancreatic cancer, intestinal cancer and endometrial carcinoma was detected by immunohistochemical staining and compared with the adjacent normal tissues. Results: The expression of Oct4 in all the five kinds of adenocarcinoma was higher than that in the adjacent normal tissues (P <0.05). The expression of Oct4 in other adenocarcinomas was not statistically different from that in paracancerous tissues ; In breast cancer and colorectal cancer, the expression of Oct4 in tumor tissues without lymph node metastasis was higher than that in lymph node metastasis (P <0.05), but not in gastric cancer, pancreatic cancer and endometrial cancer There was no correlation between the expression of Oct4 and clinical stages in the tumor tissues of 5 kinds of adenocarcinoma. Oct4 in adjacent normal tissues (except endometrial cancer) is mainly located in the cytoplasm, while in adenocarcinoma tissue is expressed in the nucleus, but also expressed in the cytoplasm. Conclusion: The expression of Oct4 may be an early event in tumorigenesis. The expression of Oct4 in different adenocarcinomas is not completely the same. The nuclear / plasma expression pattern of Oct4 may be related to its role in tumorigenesis.
