目的对2015年郴州市人感染布鲁氏菌疑似患者和病羊进行血清学和分子生物学检测,以明确诊断。方法对患者进行流行病学调查,采用血培养法对试管凝集试验检测为布鲁氏菌抗体阳性的患者和病羊血液进行荧光PCR及细菌分离培养,应用分子生物学方法和基因测序对可疑菌落进行种类鉴定。结果 68份高危及疑似布病患者血清抗体筛查13份阳性;1 679份羊血血清抗体检测57份阳性,6株分离株经荧光PCR鉴定为羊种布鲁氏菌。结论 2015年引起郴州市布病的主要病原类型为羊种布鲁氏菌,应采取综合措施防止布病在郴州流行。
Objective To investigate the serological and molecular biological tests of suspected brucellosis patients and sick sheep in Chenzhou City in 2015 in order to confirm the diagnosis. Methods The epidemiological investigation of the patients was carried out. Fluorescent PCR and bacterial isolation were performed on the blood of patients with positive brucella antibody detected by agglutination test in the tube agglutination test and blood culture. The molecular biology and gene sequencing were used to detect the suspicious bacterial colonies For species identification. Results Seventy-eight serum samples from 68 high-risk and suspected cases of brucellosis were positive for screening for serum antibodies. Six hundred and seventy-seven sera were tested positive for serum antibody against sheep serum and six isolates were identified as Brucella abortus by fluorescence PCR. Conclusion The main pathogenic species causing brucellosis in Chenzhou in 2015 is brucella brucellosis, and comprehensive measures should be taken to prevent the epidemic of brucellosis in Chenzhou.
