目的 观察HSP70反义寡脱氧核苷酸对宫颈癌细胞和卵巢癌细胞热敏感性的影响。方法计数集落形成率 ,了解宫颈癌细胞和卵巢癌细胞的热敏感性。用光镜、琼脂糖凝胶电泳及流式细胞术观察凋亡细胞。结果 经HSP70反义寡脱氧核苷酸预先处理的细胞增强了对热处理的敏感性 ,热敏感性的增加与HSP70反义寡脱氧核苷酸的处理剂量相关。用形态学、琼脂糖凝胶电泳及流式细胞术均证实经HSP70反义寡脱氧核苷酸及热处理的细胞发生了凋亡。凋亡的发生率与HSP70反义寡脱氧核苷酸的处理剂量相关。结论 HSP70反义寡脱氧核苷酸能增强宫颈癌细胞和卵巢癌细胞的热敏感性 ,增强这种热敏感性的可能机理是协同诱导细胞凋亡。
Objective To observe the effect of HSP70 antisense oligodeoxynucleotides on the thermosensitivity of cervical cancer cells and ovarian cancer cells. Methods The colony formation rate was counted to understand the thermosensitivity of cervical cancer cells and ovarian cancer cells. Apoptotic cells were observed by light microscopy, agarose gel electrophoresis and flow cytometry. Results The pretreatment of cells with HSP70 antisense oligodeoxynucleotide increased the sensitivity to heat treatment and the increase of thermosensitivity correlated with the treatment dose of HSP70 antisense oligodeoxynucleotides. Morphology, agarose gel electrophoresis and flow cytometry confirmed that HSP70 antisense oligodeoxynucleotides and heat-treated cells were apoptotic. The incidence of apoptosis correlated with the treatment dose of HSP70 antisense oligodeoxynucleotide. Conclusion HSP70 antisense oligodeoxynucleotides enhance the thermosensitivity of cervical cancer cells and ovarian cancer cells, and the possible mechanism of enhancing this thermosensitivity is synergistic induction of apoptosis.
