目的:观察通络救脑注射液对成年大鼠侧脑室室管膜下层Vimentin、胶质原纤维酸性蛋白(GFAP)阳性细胞的作用。方法:采用Wistar成年雄性大鼠,去除大鼠左侧大脑皮层血管,应用免疫组化方法观察去皮层血管对侧脑室室管膜下区(SVZ)神经干细胞增殖、分化和迁移的影响,以及通络救脑注射液的作用。结果:通过连续切片的免疫组化结果显示去皮层血管后侧脑室SVZ的增殖细胞核抗原(PCNA)、Vimentin、GFAP阳性细胞均明显增多,反应增强,表象改变显著。此种损伤刺激引起了相对静止的SVZ神经干细胞增殖、分化,给予通络救脑注射液后促进以上各种阳性细胞表达增多、增强。结论:提示通络救脑注射液可能通过上调内源性促神经生长因子的表达促进去皮层血管后成年大鼠SVZ神经干细胞增殖、分化。
OBJECTIVE: To observe the effect of Tongluo Jiunao injection on subepithelium Vimentin and glial fibrillary acidic protein (GFAP) positive cells in adult rat lateral ventricle. METHODS: Wistar adult male rats were used to remove blood vessels from the left cerebral cortex. Immunohistochemistry was used to observe the effects of cortical blood vessels on the proliferation, differentiation, and migration of neural stem cells in the subventricular zone (SVZ) of the lateral ventricle. The role of Luosuo brain injection. Results: The immunohistochemistry results of serial sections showed that the proliferating cell nuclear antigen (PCNA), Vimentin, and GFAP positive cells in the SVZ in the posterior ventricle of the cortical vessels were significantly increased, the response was enhanced, and the appearance changes were significant. This type of injury stimulation caused the proliferation and differentiation of relatively static SVZ neural stem cells, and after administration of Tongluo Jiunao Injection, it promoted the expression and enhancement of various positive cells. Conclusion: It is suggested that Tongluo Jiunao Injection may promote the proliferation and differentiation of SVZ neural stem cells in adult rats following cortical devascularization by up-regulating the expression of endogenous nerve growth factor.
