目的分析甲苯对24 h尿液蛋白定量检测的影响,探讨24 h尿液中加入甲笨防腐剂的必要性。方法在同一时间收集6 L尿液后,加入蛋白标准品,平均分为6等份,并分为2组,其中一组不加入甲苯,为对照组,另一组加入甲苯,为实验组,分别于4℃、25℃、30℃保存,在0 h、4 h、8 h、12 h、24 h、28 h、32 h不同的保存时间条件下测尿总蛋白、尿微量白蛋白、尿肌酐的量,并同时进行尿液细菌培养,18 h后进行菌落计数。结果在不同的温度下,不加防腐剂的情况下,尿总蛋白、尿微量白蛋白、尿肌酐可稳定保持32 h,且细菌菌落>10~5cfu/ml。结论用于24 h尿总蛋白、尿微量白蛋白、尿肌酐定量检测的标本可以不加入甲苯,甲苯在尿液中没有防腐效果。
Objective To analyze the effect of toluene on the quantitative determination of 24-hour urine protein and to explore the necessity of adding a stoichiometric preservative to 24-hour urine. Methods After 6 L urine was collected at the same time, the protein standard was added and the average was divided into 6 equal parts and divided into 2 groups. One group did not add toluene, the control group and the other group added toluene as experimental group, The urine samples were stored at 4 ℃, 25 ℃ and 30 ℃, respectively. Urinary total protein, urine microalbuminuria, urine microalbuminuria and urinary albumin were measured under different storage time of 0 h, 4 h, 8 h, 12 h, 24 h, 28 h and 32 h Creatinine, and urine bacterial culture at the same time, after 18 h colony count. Results Urine total protein, urine microalbuminuria and urinary creatinine were stable for 32 h at different temperatures without preservatives, and bacterial colonies> 10 ~ 5 cfu / ml. Conclusion The 24 h urine total protein, urine microalbuminuria, urinary creatinine quantitative detection of specimens can be added without toluene, toluene in the urine without preservative effect.
