目的探讨脐血源间充质干细胞(mesenchymal stem cells derived from umbilical cord blood,UCB-MSCs)联合外源性细胞因子对脐血单个核细胞(umbilical cord blood mononuclear cells,UCB-MNCs)体外扩增的支持作用。方法从脐血中培养出UCB-MSCs,检测其表面抗原,并以此作为滋养层细胞,将UCB-MNCs接种于无血清培养体系中培养10d,检测有核细胞总数(MNCs)、CD34+、CD133+细胞数、集落形成单位数(CFU)和(G2+M+S)期细胞含量的变化。结果从脐血分离、培养出UCB-MSCs,稳定表达CD29、CD105和CD44,不表达CD34和CD133。外源性细胞因子及UCB-MSCs均支持UCB-MNCs的扩增,但以细胞因子联合UCB-MSCs组效果最好(P<0.05)。结论从脐血中能成功分离培养出间充质干细胞,并完成细胞表型的初步鉴定。外源性细胞因子联合UCB-MSCs可有效扩增UCB-MNCs。
Objective To investigate the in vitro expansion of umbilical cord blood derived mononuclear cells (UCB-MNCs) derived from mesenchymal stem cells derived from umbilical cord blood (UCB-MSCs) and exogenous cytokines Support role. Methods UCB-MSCs were cultured from umbilical cord blood and the surface antigens were detected. As a feeder cell, UCB-MNCs were inoculated into serum-free culture medium for 10 days. The number of nucleated cells (MNCs), CD34 +, CD133 + Cell number, colony forming unit number (CFU) and (G2 + M + S) phase cell content changes. Results UCB-MSCs were isolated from umbilical cord blood and stably expressed CD29, CD105 and CD44 but not CD34 and CD133. Both exogenous cytokines and UCB-MSCs supported the expansion of UCB-MNCs, but the combination of cytokines and UCB-MSCs had the best effect (P <0.05). Conclusion The mesenchymal stem cells can be successfully isolated and cultured from umbilical cord blood, and the initial identification of the cell phenotype is completed. Exogenous cytokines combined with UCB-MSCs can effectively amplify UCB-MNCs.
