目的构建含有K10Q、R30I、S165W及K10Q/R30I/S165W4种t PA突变体。方法采用PCR重叠延伸法进行突变体的构建。结果成功地构建了上述4种t PA突变体克隆质粒 ,测序结果表明构建完全正确。结论PCR重叠延伸法为构建突变体的可靠快捷的方法。
Objective To construct t PA mutants containing K10Q, R30I, S165W and K10Q / R30I / S165W. Methods The mutant was constructed by PCR overlap extension method. Results The cloning plasmids of the four t PA mutants were successfully constructed and the sequencing results showed that the construction was correct. Conclusion PCR overlap extension method is a reliable and efficient method to construct mutants.
