目的研究反义VEGF基因对人胶质细胞瘤BT325生长的抑制作用及由此引起的细胞凋亡。方法构建含VEGF反义基因的真核表达载体pc DNA3.1/anti-VEGF(实验组),将其转染人胶质细胞瘤BT325,ELISA检测VEGF的表达。采用软琼脂集落生成,MTT以及电子显微镜观察胶质细胞瘤的增殖及凋亡等变化。结果与对照组相比,实验组VEGF的表达显著降低,几乎完全被抑制。与空载体组细胞集落生成数(21)相比,实验组细胞集落生成数(2)明显减少,胶质细胞瘤的生长受到明显的抑制,抑制率为38.23%,且电镜下实验组细胞出现明显的凋亡形态。结论反义VEGF基因通过抑制VEGF的表达抑制人胶质细胞瘤的增殖,促进细胞凋亡,发挥抗癌作用。
Objective To study the inhibitory effect of antisense VEGF gene on the growth of human glioblastoma BT325 cells and the resulting apoptosis. Methods The eukaryotic expression vector pcDNA3.1 / anti-VEGF (experimental group) containing VEGF antisense gene was constructed and transfected into human glioblastoma cell line BT325. The expression of VEGF was detected by ELISA. Soft agar colony formation, MTT and electron microscopy were used to observe the changes of glioblastoma proliferation and apoptosis. Results Compared with the control group, the expression of VEGF in the experimental group was significantly reduced and almost completely inhibited. Compared with the empty vector group, the number of colony-forming cells (2) in the experimental group was significantly decreased compared with that in the empty vector group (21), the growth of the glioma was significantly inhibited with the inhibition rate of 38.23%, and the cells in the experimental group appeared under the electron microscope Obvious apoptotic morphology. Conclusion The antisense VEGF gene can inhibit the proliferation of human glioma by inhibiting the expression of VEGF, promote apoptosis and exert its anticancer effect.
