目的:探讨miR-155表达水平在链脲佐菌素(STZ)诱导糖尿病肾病(DN)大鼠及大鼠肾系膜细胞中是否存在同样的变化。方法:将雄性清洁SD大鼠8只随机分成实验组(DN组,n=4)和对照组(NC组,n=4),DN组大鼠腹腔注射足量STZ诱导DN大鼠,观察血糖及肾脏结构变化,用real-time PCR检测肾脏miR-155表达水平;大鼠肾系膜细胞分为高糖培养组和正糖培养组,培养24 h、48 h、72 h,用real-time PCR检测miR-155表达水平。结果:与对照组相比,DN组大鼠肾脏miR-155表达水平下降(P<0.05);高糖培养的大鼠肾系膜细胞miR-155表达水平较正糖培养的表达水平下降(P<0.05),于48 h表达水平最低。结论:miR-155在高糖培养大鼠肾系膜细胞表达下降。
AIM: To investigate whether miR-155 expression is the same in streptozotocin (STZ) -induced diabetic nephropathy (DN) rats and rat mesangial cells. Methods: Eight male SD rats were randomly divided into experimental group (DN group, n = 4) and control group (NC group, n = 4). DN rats were injected intraperitoneally with adequate STZ to induce DN rats. The renal mesangial cells were divided into high glucose group and normal glucose group and cultured for 24 h, 48 h, 72 h. Real-time PCR Detection of miR-155 expression levels. Results: Compared with the control group, the expression of miR-155 in the kidney of DN group was decreased (P <0.05); the expression of miR-155 in mesangial cells of high glucose group was lower than that of the normal glucose group <0.05), the lowest expression level at 48 h. Conclusion: The expression of miR-155 is decreased in mesangial cells cultured in high glucose.
