目的,建立用微量稀释法测定皮肤癣菌最低抑菌浓度(MIC)的方法学,为该类实验的标准化打下基础。方法:用96孔酶标板做药敏试验载体,10级倍比稀释药液,测定30株皮肤癣菌临床分离株对三唑类新药伊曲康唑和氟康唑的敏感性。结果:30株皮肤癣菌对伊曲康唑和氟康唑的MIC范围分别为≤0.06~4μg/ml和1~32μg/ml,均值分别为0.32μg/ml和7.76μg/ml.结论:用微量稀释法测定皮肤癣菌MIC初步显示了用药量少、重复性好、结果较可靠等优点。
Objective To establish a method for the determination of minimum inhibitory concentration (MIC) of dermatophytes by the method of microdilution, laying a foundation for the standardization of such experiments. Methods: The drug susceptibility test vector and 10-fold dilution of 96-well microplate were used to determine the susceptibility of 30 dermatophytes isolates to itraconazole and fluconazole. Results: The MICs of 30 dermatophytes against itraconazole and fluconazole were ≤0.06 ~ 4μg / ml and 1 ~ 32μg / ml respectively with the mean values of 0.32μg / ml and 7.76μg / ml respectively . Conclusion: The determination of dermatophytes MIC by microdilution shows the advantages of less dosage, better repeatability and more reliable results.
