目的研究细胞因子(TGFβ1,CTGF和IFN-γ)对系统性硬皮病具有不同胶原合成能力的克隆成纤维细胞胶原代谢的影响。方法采用实时荧光定量RT-PCR法检测细胞因子(TGFβ1,CTGF和IFN-γ)对系统性硬皮病具有不同胶原合成能力的克隆成纤维细胞Ⅰ型前胶原和MMP-1 mRNA水平的影响情况。结果 TGFβ1(5ng/mL)作用48h仅使具有低胶原合成能力的系统性硬皮病克隆成纤维细胞的Ⅰ型前胶原mRNA水平上升(1.20±0.50vs0.04±0.04,P=0.001);CTGF(20ng/mL),IFN-γ(100ng/mL)作用48h使系统性硬皮病克隆成纤维细胞MMP-1 mRNA水平下降(F=6.221,P=0.034;F=53.505,P=0.000),且具有高胶原合成能力的克隆成纤维细胞MMP-1 mRNA水平下降幅度大(F=8.817,P=0.016;F=8.092,P=0.019)。结论系统性硬皮病具有不同胶原合成能力的克隆成纤维细胞Ⅰ型前胶原和MMP-1 mRNA水平对细胞因子的反应存在异质性。
Objective To investigate the effects of cytokines (TGFβ1, CTGF and IFN-γ) on the collagen metabolism of cloned fibroblasts with different collagen synthesis ability in systemic scleroderma. Methods The effects of cytokines (TGFβ1, CTGF and IFN-γ) on the expression of type Ⅰ procollagen and MMP-1 mRNA of clonal fibroblasts with differential collagen synthesis in systemic scleroderma were detected by real-time fluorescent quantitative RT-PCR . Results TGFβ1 (5ng / mL) increased the mRNA level of type Ⅰ procollagen (1.20 ± 0.50vs0.04 ± 0.04, P = 0.001) in systemic scleroderma clot fibroblasts with low collagen synthesis ability only for 48h; CTGF (F = 6.221, P = 0.034; F = 53.505, P = 0.000) in the group of systemic scleroderma clonal fibroblasts after treated with IFN-γ (20ng / mL) The mRNA level of MMP-1 in clonal fibroblasts with high collagen synthesis ability decreased greatly (F = 8.817, P = 0.016; F = 8.092, P = 0.019). Conclusions There is heterogeneity in the response of cytokines to type Ⅰ procollagen and MMP-1 mRNA in clone fibroblasts with different sclerotic ability of scleroderma.
