目的:探讨S-腺苷蛋氨酸(思美泰)对大鼠肝缺血再灌注损伤后清蛋白(ALB)mRNA表达的影响。方法:将72只SD大鼠随机均分为假手术组,肝缺血再灌注组(IR组),肝缺血再灌注+腺苷蛋氨酸干预组(SAM组)。IR组和SAM组均以肝脏缺血20 min后再灌注的方式造模。SAM组于术后1 h腹腔注射思美泰100 mg/(kg.d),假手术组和IR组予以等体积生理盐水。各组分别于术后1,4,7,10 d处死6只大鼠,用实时定量PCR方法测定肝组织ALB mRNA的含量。结果:术后第1天SAM组和IR组ALB mRNA相对表达量较假手术组明显降低(均P<0.05),但两组间差异无统计学意义(P>0.05);从术后第4天开始,SAM组ALB mRNA的相对表达量明显高于IR组(P<0.05),且至术后第10天明显超过假手术组(P<0.05)。结论:腺苷蛋氨酸能提高肝缺血再灌注损伤后ALB mRNA的合成,增强肝脏的合成功能。
Objective: To investigate the effect of S-adenosylmethionine (Smectite) on the expression of albumin (ALB) mRNA after hepatic ischemia-reperfusion injury in rats. Methods: Seventy-two SD rats were randomly divided into sham operation group, IR group, IR group and SAM group. The IR and SAM groups were all established by reperfusion after 20 min of hepatic ischemia. The SAM group was injected with Smectite 100 mg / (kg.d) intraperitoneally 1 h after operation, and the rats in sham operation group and IR group were given equal volume of normal saline. Six rats were sacrificed on the 1st, 4th, 7th and 10th postoperative day in each group, and the content of ALB mRNA in liver tissue was determined by real-time PCR. Results: On the first postoperative day, the relative expression of ALB mRNA in SAM group and IR group was significantly lower than that in sham operation group (all P <0.05), but there was no significant difference between the two groups (P> 0.05) The relative expression level of ALB mRNA in SAM group was significantly higher than that in IR group (P <0.05), and was significantly higher than that in sham operation group on the 10th day after operation (P <0.05). CONCLUSION: Adenosine methionine can increase ALB mRNA synthesis after hepatic ischemia-reperfusion injury and enhance hepatic synthesis.
