为了了解偶发分枝杆菌的超微结构 ,并对其结构与功能关系进行初步探讨 ,我们采用负染色法和超薄切片法等电镜技术进行观察。其负染色观察表明 ,菌体为长杆或短杆菌 ,多呈直杆或微曲杆状 ,未见鞭毛 ,大小 0 .4~ 0 .5× 1.5~ 3.5 μm。可见二分裂相 ,菌体有的呈分枝状排列。超薄切片观察可见 ,菌体纵切面或斜切面呈长杆或短杆状 ,横斜切面多见呈圆形、椭圆形 ,可见一些菌体呈曲棍球拍样。菌体为革兰氏阴性杆菌结构 ,细胞壁较薄 ,细胞膜可见 ,胞壁外有时可见细绒样物。菌体胞质中常充满核糖体 ,并常见一个至十几个高电子密度的小颗粒。可见菌体近末端有电子密度较浅的拟核区 ,偶见有数层片层状结构
In order to understand the ultrastructure of Mycobacterium fortuitum, and its structure and function of the preliminary study, we use negative staining and ultra-thin section method such as electron microscopy observation. Negative staining showed that the cells were long or short bacteria, mostly straight or slightly curved rods, no flagella, the size of 0.4 ~ 0.5 × 1.5 ~ 3.5 μm. Seen the two split phase, some bacteria were branched arrangement. Ultrathin sections can be seen, the cell longitudinal section or oblique section was a long rod or short rod-like, horizontal oblique surface more common round, oval, showing that some bacteria hockey shoot like. Gram-negative bacilli bacterial cell structure, the cell wall is thin, the cell membrane can be seen, sometimes seen outside the cell wall velvet samples. Fungal cytoplasm is often filled with ribosomes, and a common one to a dozen high electron density of small particles. Can be seen near the end of bacterial cells with electron density shallow nuclear region, and occasionally a few lamellar layered structure
