目的 :探讨内毒素休克大鼠肝窦内皮细胞的损伤及活血化瘀药的保护作用。方法 :选健康 Wistar大鼠 4 0只 ,随机平分为正常对照组 (NS)、模型组 (L PS)、地塞米松组 (Dex)和活血化瘀药组 (AMRS)。电镜观察各组大鼠肝窦内皮细胞的超微结构变化 ,并比较其损伤程度。结果 :L PS组肝窦内皮细胞功能不良 ,溶解、消失、间隙扩大 ,枯否细胞增生及窦腔内白细胞黏附 ,纤维素凝结 ,病变最为严重 ;Dex组的变化均较轻 ;而 AMRS组的变化最为轻微 ,内皮细胞结构趋于正常 ,未见窦腔内白细胞黏附 ,纤维素凝结。结论 :由内毒素诱导的肝窦内皮细胞超微结构损伤是严重的 ,活血化瘀药在内毒素休克中对内皮细胞具有明显的保护作用。
Objective : To investigate the damage of liver sinusoidal endothelial cells and the protective effect of blood circulation and stasis drugs in rats with endotoxin shock. Methods: Forty healthy Wistar rats were randomly divided into normal control group (NS), model group (L PS), dexamethasone group (Dex) and Huoxuehuayu medicine group (AMRS). Electron microscopy was used to observe the ultrastructural changes of sinusoidal endothelial cells in each group and to compare the degree of damage. Results: The sinusoidal endothelial cells in LPS group had poor function, dissolved, disappeared, gaps expanded, Kupffer cell proliferation and leucocyte adhesion in sinusoids, cellulose coagulation, and lesions were the most serious; the changes in Dex group were lighter; while in AMRS group, The change was slightest, the structure of endothelial cells tended to be normal, and there was no adhesion of white blood cells in the sinus cavity and cellulose coagulation. Conclusion: Endotoxin-induced morphological damage of liver sinusoidal endothelial cells is serious, and blood circulation and phlegm-removing drugs have a significant protective effect on endothelial cells in endotoxin shock.
