目的研究活动性结核患者单核细胞中B淋巴细胞诱导成熟蛋白1(Blimp-1)的mRNA水平与结核分枝杆菌(MTB)潜伏感染者和健康者相比是否发生改变。方法收集活动性结核患者、结核菌潜伏感染者和和健康者的抗凝外周血,从外周血中分离并纯化单核细胞,荧光定量PCR检测单核细胞中Blimp-1 mRNA的水平。而后分别用MTB的H37Rv菌株全菌裂解液和ESAT-6/CFP-10的抗原多肽刺激健康者的单核细胞,荧光定量PCR检测刺激前后单核细胞中Blimp-1 mRNA的水平。结果与健康者相比,活动性结核患者、潜伏感染者单核细胞Blimp-1的mRNA水平显著降低;与未刺激的单核细胞相比,全菌裂解液刺激后的单核细胞Blimp-1的mRNA水平显著降低,而ESAT-6/CFP-10抗原多肽刺激后的单核细胞中Blimp-1的mRNA水平无显著变化。结论活动性结核患者和MTB潜伏感染者单核细胞Blimp-1的mRNA水平降低,MTB全菌裂解液刺激的人单核细胞中Blimp-1的mRNA水平降低。
Objective To investigate whether the mRNA level of B lymphocyte-induced mature protein 1 (Blimp-1) in patients with active tuberculosis (TB) is changed compared with those in latent infection and healthy subjects with Mycobacterium tuberculosis (MTB). Methods Peripheral blood mononuclear cells (PBMCs) were collected from patients with active tuberculosis, latent infection with tuberculosis and healthy individuals. Mononuclear cells were isolated and purified from peripheral blood. Fluorescence quantitative PCR was used to detect the level of Blimp-1 mRNA in mononuclear cells. Then mononuclear cells of healthy individuals were stimulated by whole cell lysate of MTB strain H37Rv and antigenic peptide of ESAT-6 / CFP-10, respectively. Fluorescence quantitative PCR was used to detect the level of Blimp-1 mRNA in mononuclear cells before and after stimulation. Results The mRNA level of Blimp-1 in active tuberculosis patients and latent infection patients was significantly lower than that in healthy subjects. Compared with unstimulated monocytes, the expression of Blimp-1 in monocytes stimulated by whole bacteria lysate MRNA levels of Blimp-1 mRNA were significantly decreased in monocytes stimulated with ESAT-6 / CFP-10 antigen peptide. Conclusions The mRNA level of Blimp-1 in mononuclear cells of patients with active tuberculosis and latent infection of MTB is decreased, and the mRNA level of Blimp-1 in human monocytes stimulated by MTB whole cell lysate is decreased.
