目的:研究血管紧张素Ⅱ受体AT_1R和AT_2R在肾上腺产醛固酮腺瘤(APA)中的表达。方法:通过逆转录聚合酶链反应(RT-PCR)检测50例APA及瘤旁组织和12例正常肾上腺组织中AT_1R和AT_2R mRNA表达,采用免疫组织化学染色方法检测组织石蜡切片中AT_1R和AT_2R蛋白表达情况。结果:AT_1R在腺瘤、瘤旁及正常肾上腺组织中的表达无明显差异。AT_2R在APA组织中的表达低于正常肾上腺组织(P<0.05)。将AT_2R mRNA表达量与患者临床数据作相关性分析,提示AT_2R的表达与患者血浆醛固酮浓度负相关(r=-0.467,P<0.05),与血浆肾素活性正相关(r=0.604,P<0.05)。结论:AT_2R表达下调可能与肾上腺产醛固酮腺瘤原发性醛固酮增多症的发病相关。
Objective: To investigate the expression of angiotensin Ⅱ receptor AT_1R and AT_2R in adrenal aldosterone producing adenoma (APA). Methods: The expressions of AT1R and AT2R mRNA in 50 cases of APA, adjacent normal tissue and 12 cases of normal adrenal tissue were detected by reverse transcriptase polymerase chain reaction (RT-PCR). The expression of AT1R and AT2R in paraffin sections was detected by immunohistochemical staining Express the situation. Results: There was no significant difference in the expression of AT_1R between adenoma, para-tumor and normal adrenal tissue. The expression of AT_2R in APA tissues was lower than that in normal adrenal tissues (P <0.05). Correlation analysis showed that the expression of AT_2R mRNA was negatively correlated with plasma aldosterone concentration (r = -0.467, P <0.05), and positively correlated with plasma renin activity (r = 0.604, P < 0.05). Conclusion: The down-regulation of AT_2R expression may be related to the pathogenesis of primary aldosteronism of adrenal aldosterone-producing adenoma.
