新近研究发现,神经生长因子(NGF)对非神经系统也具有重要的功能效应。为探讨NGF与骨髓造血组织的关系,本实验制备地高辛标记的人βNGFcDNA探针,通过原位杂交技术,检测正常人骨髓NGFmRNA的分布。实验显示,βNGFcDNA探针与其互补的mRNA杂交反应信号存在于巨核细胞系和组织嗜碱细胞,杂交反应产物呈紫蓝色颗粒,分布于阳性细胞胞浆中。实验结果提示,NGF基因在骨髓造血组织也有表达,可能参与造血功能的调节。本实验制备的地高辛标记βNGFcDNA探针灵敏度高达0.1pg/ml,且特异性强、分辨力高、重复性好,是研究NGF荃因表达和调控的有力工具。
Recent studies have found that nerve growth factor (NGF) also has important functional effects on the non-nervous system. In order to explore the relationship between NGF and bone marrow hematopoietic tissue, digoxin-labeled human βNGF cDNA probe was prepared in this study to detect the distribution of NGF mRNA in normal human bone marrow by in situ hybridization. Experiments show that, βNGF cDNA probe and its complementary mRNA hybridization reaction signal exists in the megakaryocytic and tissue basophils, the hybridization reaction product was purple-blue particles, located in the cytoplasm of positive cells. Experimental results suggest that NGF gene is also expressed in bone marrow hematopoietic tissue may be involved in the regulation of hematopoietic function. The digoxin-labeled βNGF cDNA probe prepared in this experiment has a sensitivity of 0.1 pg / ml and strong specificity, high resolution and good reproducibility. It is a powerful tool to study the expression and regulation of NGF.
