目的:探讨与细胞增殖相关的端粒酶逆转录酶TERT、增殖细胞核抗原(PCNA)和凋亡抑制蛋白Bcl-2在外伤性PVR视网膜前膜形成中的动态变化及其相关性。方法:S-P法对外伤性PVR大鼠模型视网膜前膜做TERT、PCNA、Bcl-2免疫组织化学染色和HE染色,染色结果行图像分析。结果:伤后PCNA蛋白表达阳性细胞率和平均光密度值出现先升高后降低的动态变化,14d组阳性率最高,与7d组和28d组比较有显著性差异。伤后TERT蛋白和Bcl-2表达阳性细胞率和平均光密度值出现先升高后稍降低的动态变化,14d组和21d组阳性率最高,与7d组比较有显著性差异,TERT、PCNA和Bcl-2蛋白表达之间有显著相关性(P ≤0.01)。结论:TERT、Bcl-2参与外伤性PVR视网膜前膜增殖细胞的生长调控,与细胞增殖的动态变化呈高度相关性。
Objective: To investigate the dynamic changes of telomerase reverse transcriptase (TERT), proliferating cell nuclear antigen (PCNA) and apoptosis-inhibitory protein (Bcl-2) in the development of traumatic PVR retinal membrane and their correlation. Methods: The TPR, PCNA and Bcl-2 immunohistochemical staining and HE staining were performed on the anterior retinal membrane of traumatic PVR rat model by S-P method. The results of staining were analyzed by image analysis. Results: The positive cell rate and average optical density of PCNA protein increased firstly and then decreased, the highest positive rate was found in 14d group, which was significantly different from that in 7d group and 28d group. The positive rates of TERT protein and Bcl-2 positive cells and the average optical density of the injured rats increased firstly and then decreased slightly, the positive rates of 14th and 21d groups were the highest, which were significantly different from those of 7d group There was a significant correlation between Bcl-2 protein expression (P ≤ 0.01). CONCLUSION: TERT and Bcl-2 are involved in the regulation of proliferation of proliferating cells in the pre-retinal membrane of traumatic PVR and are highly correlated with the dynamic changes of cell proliferation.
