目的:分析肝细胞肝癌与正常肝组织中RICH2蛋白表达之间的关系,探讨RICH2在肝细胞肝癌发生发展中的作用机理。方法:选取19例肝细胞肝癌患者作为研究对象,用免疫组织化学方法检测肝癌组织和癌旁正常组织中RICH2的表达情况并采用Friedrich免疫反应评分法作为判定标准。采用非参数Wilcoxon秩和检验对RICH2在肝细胞肝癌和正常组织中的评分结果进行统计学分析。结果:19例HCC病例癌旁正常组织中RICH2表达阳性率为100%(19/19),其中RICH2表达强阳性(+++)5例(26.3%),RICH2表达弱阳性(+)和中阳性(++)分别为3例(15.8%)和11例(57.9%);而HCC组织除了两例肝硬化组织未取到癌组织外,17例病例中RICH2表达阳性率仅为11.8%(2/17),表达为+和++各占50%。肝癌组织中RICH2的表达水平显著低于癌旁组织。结论:RICH2有可能通过某种或者其中几种因素作用失活,作用于信号通路,进而促进肝细胞肝癌的发生发展。研究其作用机理,可为临床诊断、治疗及预后评估提供理论依据。
OBJECTIVE: To analyze the relationship between the expression of RICH2 protein in hepatocellular carcinoma and normal liver tissue and to explore the mechanism of RICH2 in the occurrence and development of hepatocellular carcinoma. Methods: Totally 19 patients with hepatocellular carcinoma were enrolled in this study. The expression of RICH2 in hepatocellular carcinoma (HCC) and adjacent normal tissues was detected by immunohistochemical method and the Friedrich immune response score was used as the criterion. Non-parametric Wilcoxon rank-sum test was used to analyze the scores of RICH2 in hepatocellular carcinoma and normal tissues. Results: The positive rate of RICH2 expression was 100% (19/19) in the adjacent normal tissues of 19 cases of HCC, including 5 cases (26.3%) with RICH2 positive expression (+++), weak positive (+) and Positive (++) were 3 cases (15.8%) and 11 cases (57.9%) respectively. The positive rate of RICH2 expression was only 11.8% (17.8%) in 17 cases of HCC tissues except two cirrhosis tissues 2/17), expressed as + and ++ each 50%. The expression level of RICH2 in HCC tissues was significantly lower than that in paracancerous tissues. CONCLUSION: RICH2 may be inactivated by one or several of these factors and act on the signal pathways to promote the development of hepatocellular carcinoma. To study its mechanism of action may provide a theoretical basis for clinical diagnosis, treatment and prognosis evaluation.
