目的通过体外药效评价重楼皂苷Ⅰ对肺癌循环肿瘤细胞(circulating tumor cells,CTC)增殖、凋亡的影响。方法分别用不同浓度的重楼皂苷Ⅰ干预CTC细胞24 h、48 h、72 h后,采用CCK-8法检测细胞增殖情况,采用流式细胞术和激光共聚焦显微镜分析CTC凋亡与周期情况。结果重楼皂苷Ⅰ能够显著抑制CTC增殖并具有浓度依赖性,诱导CTC细胞核形态发生明显改变,使凋亡细胞的比例显著升高,并将CTC细胞的周期阻滞在G0/G1期。结论重楼皂苷Ⅰ通过诱导CTC凋亡,抑制细胞从G0/G1期向S期转化,从而发挥抗肿瘤作用。
Objective To evaluate the effect of polysaccharide Ⅰ on the proliferation and apoptosis of circulating tumor cells (CTCs) in lung cancer cells in vitro. Methods CTC cells were treated with different concentrations of Polysaccharide Ⅰ for 24 h, 48 h and 72 h, respectively. The proliferation of CTCs was detected by CCK-8 assay. The apoptosis and cycle of CTCs were analyzed by flow cytometry and laser confocal microscopy . Results polysaccharide saponins Ⅰ can significantly inhibit CTC proliferation in a concentration-dependent manner, CTC nuclear morphogenesis induced significantly changed, the proportion of apoptotic cells was significantly increased, and the cycle of CTC cells in G0 / G1 phase. Conclusions Saponin Ⅰ exerts antitumor effect by inducing CTC apoptosis and inhibiting the transformation of cells from G0 / G1 phase to S phase.
