评价外周血淋巴细胞凋亡在肾小球肾炎(GN)发病中的作用。方法用免疫组化、荧光显微镜及电子显微镜对GN进行病理分型,采用流式细胞术测定不同病理类型GN患者外周血单个核细胞(PBMC)Bcl-2、Fas表达水平。结果各型GNPBMCBcL-2表达水平均高于对照组(P<0.01),Fas均低于对照组(P<0.01);增殖型GN与非增殖型GN及增殖型GN中IgAN、MPGN、HSPN新月体组与非月体组间,Bcl-2、Fas表达水平存在显著差异,前者Bcl-2高于后者(P<0.01),Fas低于后者(P<0.01)。结论Bcl-2过量表达,Fas系统功能不良,活化PBMC增多,导致肾小球肾炎。Bcl-2、Fas表达水平与GN病理类型及临床预后有一定联系。
To evaluate the role of peripheral blood lymphocyte apoptosis in the pathogenesis of glomerulonephritis (GN). Methods Immunohistochemistry, fluorescence microscopy and electron microscopy were used to determine the pathological type of GN. Flow cytometry was used to determine the expression of Bcl-2 and Fas in peripheral blood mononuclear cells (PBMCs) from patients with different pathological types of GN. Results The expression of GNPBMCBcL-2 in each group was significantly higher than that in the control group (P <0.01), Fas was significantly lower than that in the control group (P <0.01), while in the proliferating GN, non-proliferating GN and IgAN, There were significant differences in the expression of Bcl-2 and Fas between MPGN and HSPN crescent group and non-crescent group, the former was higher than the latter (P <0.01) .01). Conclusion Overexpression of Bcl-2, dysfunction of Fas system and increase of activated PBMC lead to glomerulonephritis. The expression of Bcl-2 and Fas is related to the pathological type of GN and clinical prognosis.
