目的在细胞水平研究高血压病人动脉血管平滑肌细胞(ASMC)的生长特性和体外血管活性物质对ASMC增殖作用的影响。方法应用复合酶法进行高血压病人ASMC分离,并用人胰岛素样生长因子I(HILGF-I)干预以观察人ASMC的增殖情况。结果经低浓度的HILGF-I作用,高血压病人ASMC呈典型的“谷”与“峰”样生长,经历了10多个月传代培养至35代。发现该细胞生长分裂活性活跃,与人胎儿脐动脉增殖活性相似。结论复合酶法能使高血压病人的ASMC的静止期缩短,传代后生长特征稳定性强,高血压病人体外培养ASMC的增殖有赖于HILGF-1的存在。
Objective To study the growth characteristics of arterial smooth muscle cells (ASMCs) and the effects of extravascular vasoactive substances on the proliferation of ASMCs in hypertensive patients at the cellular level. Methods The compound enzyme method was used to separate the ASMC from patients with hypertension and intervention with human insulin-like growth factor I (HILGF-I) to observe the proliferation of human ASMC. Results With low concentration of HILGF-I, the ASMCs of hypertensive patients showed typical “valley” and “peak” -like growth and had undergone subcultures for more than 10 months to 35 passages. The cell was found to be active in growth and division, similar to human umbilical artery proliferation activity. Conclusions The compound enzyme method can shorten the quiescence of ASMC in hypertensive patients and the growth characteristics after the passage are stable. The proliferation of ASMC cultured in hypertensive patients depends on the existence of HILGF-1.
