为探讨万寿菊属远缘杂交种与双亲的遗传信息传递规律和杂交种形态表征与双亲遗传相似性的关系,本研究以筛选出的21条引物对万寿菊雪域一号及其亲本的基因组DNA进行了ISSR-PCR扩增,分析了种间杂种与亲本间的遗传相似性及其多态性差异。结果表明,总的扩增带数目为287条,其中多态性条带201条,占扩增谱带数的70.03%;雪域一号与父本雪域共显性条带共计64条,雪域一号与母本(2-2不育基因型)共显性条带共计31条,杂交种与父本的遗传相似系数为0.626 9,与母本的遗传相似系数为0.333 3,表现出杂交种与父本的共显性条带数和遗传相似系数均大于母本,间接揭示杂交种在形态特征和亲缘性方面更多倾向于父本。研究结果也证实了ISSR分子标记能够用于万寿菊远缘杂交种与亲本遗传关系及DNA多态性检测。
In order to explore the relationship between the genetic information transmission and the morphological characterization of hybrids and the genetic similarity of parents in distant hybrids and parents of Tageta spp., 21 pairs of primers were selected to screen the genomes of Xiyuan No.1 and its parents DNA was amplified by ISSR-PCR and analyzed the genetic similarity and polymorphism differences between interspecific hybrids and their parents. The results showed that the total number of amplified bands was 287, of which 201 polymorphic bands accounted for 70.03% of the number of amplified bands; a total of 64 co-dominant bands in snowy and paternal snowy areas, There were 31 co-dominant bands in Xueyue 1 and maternal (2-2 sterile genotypes), the genetic similarity coefficient between hybrids and male parents was 0.626 9, and the genetic similarity coefficient with female parents was 0.333 3. The total number of dominant bands and genetic similarity coefficients of hybrids and paternal parents were greater than that of female parents, which indirectly revealed that hybrids tended to be more paternal in their morphological characteristics and relatives. The results also confirmed that the ISSR molecular markers could be used for the genetic relationship and DNA polymorphism of Marigold distant hybrids and their parents.
