目的探讨过氧化物酶体增殖激活物受体γ(PPAR-γ)的配体曲格列酮(TGZ)对大鼠垂体腺瘤GH3细胞系白细胞分化抗原147(CD147)、转化生长因子β1(TGF-β1)和基质金属蛋白酶9(MMP-9)表达的影响。方法GH3细胞分为空白对照组和实验组(TGZ浓度分别为1×10-3、1×10-4和1×10-5mol/L)。TGZ作用GH3细胞48 h后,分别采用Western blot和荧光定量PCR检测各组细胞中CD147、TGF-β1、MMP-9蛋白和mRNA的表达。结果各组细胞CD147、TGF-β1、MMP-9蛋白和mRNA的表达与空白对照组相比,均有统计学差异(P均<0.05),而且TGZ浓度越高,CD147、TGF-β1、MMP-9蛋白和mRNA的表达量越低。结论 PPAR-γ配体TGZ能降低CD147、TGF-β1、MMP-9的表达,并可能通过该作用减弱GH3细胞的侵袭力。
Objective To investigate the effect of TGZ, a ligand of PPAR-γ, on the expression of CD147, TGFβ1 TGF-β1) and matrix metalloproteinase 9 (MMP-9) expression. Methods GH3 cells were divided into blank control group and experimental group (TGZ concentrations of 1 × 10-3, 1 × 10-4 and 1 × 10-5 mol / L, respectively). After GHZ cells were treated with TGZ for 48 h, the expressions of CD147, TGF-β1 and MMP-9 in each group were detected by Western blot and real-time PCR respectively. Results The expressions of CD147, TGF-β1, MMP-9 protein and mRNA in each group were significantly different from those in the blank control group (all P <0.05). The higher the TGZ concentration, the higher the CD147, TGF- -9 protein and mRNA expression of the lower. Conclusion PPAR-γ ligand TGZ can reduce the expression of CD147, TGF-β1 and MMP-9, and may reduce the invasiveness of GH3 cells through this effect.
