目的:观察不同浓度葡萄糖和高糖波动对具有人成骨细胞表型特征的MG63细胞增殖、细胞周期及凋亡的影响,探讨糖尿病性骨质疏松症的发病机制。方法:用不同浓度葡萄糖(5.5、33.3和5.5、33.3mmol/L交替)分别刺激培养的MG63细胞24h,MTT比色分析法测定细胞增殖,流式细胞仪检测细胞周期及凋亡率。结果:①高糖及高糖波动可抑制MG63细胞的增殖,与高糖组相比,高糖波动组的抑制效应更加明显;②高糖及高糖波动可阻滞MG63细胞周期,使G1期细胞比例增加,S期比例减少,诱导细胞凋亡。高糖波动组作用更加明显。结论:高糖波动可抑制成骨细胞增殖,阻滞其细胞周期,诱导细胞凋亡。推测高糖波动可能通过对成骨细胞增殖及细胞周期、细胞凋亡的影响而参与糖尿病性骨质疏松症的发生发展。
OBJECTIVE: To observe the effects of different concentrations of glucose and high glucose on the proliferation, cell cycle and apoptosis of MG63 cells with phenotypic characteristics of human osteoblasts and to explore the pathogenesis of diabetic osteoporosis. Methods: The cultured MG63 cells were stimulated with different concentrations of glucose (5.5,33.3 and 5.5,33.3mmol / L, respectively) for 24h. MTT colorimetric assay was used to determine the cell proliferation. Flow cytometry was used to detect cell cycle and apoptosis rate. The results showed that: (1) high glucose and high glucose fluctuations can inhibit the proliferation of MG63 cells, compared with the high glucose group, the inhibition effect of high glucose fluctuation group is more obvious; ② high glucose and high glucose fluctuations can block MG63 cell cycle, G1 The proportion of cells increased, the proportion of S phase decreased, induced apoptosis. High sugar fluctuations group more obvious effect. Conclusion: Fluctuation of high glucose can inhibit osteoblast proliferation, arrest its cell cycle and induce apoptosis. Speculated that high glucose fluctuations may be through the proliferation of osteoblasts and cell cycle, apoptosis involved in the development of diabetic osteoporosis.
