探讨烹调油烟细颗粒物对胎鼠肺泡Ⅱ型上皮细胞(AECⅡ)存活率的影响及其遗传毒性。用不同浓度PM2.5对细胞染毒12 h、24 h、48 h,MTT法测细胞存活率;彗星实验检测细胞尾长、尾部DNA百分比、尾矩和Olive尾矩;ELISA试剂盒检测细胞内PAH-DNA加合物的含量。结果显示:(1)PM2.5作用于AECⅡ12 h、24 h、48 h后,产生明显细胞毒性,呈剂量-反应和时间-反应关系;(2)染毒24 h,与低浓度组相比,高浓度组的尾长、尾部DNA百分比、尾矩和Olive尾矩有明显增加(P<0.001);与低浓度染毒组相比,高浓度染毒组的PHA-DNA加合物明显的增加(P<0.05)。提示PM2.5对AECⅡ具有明显的遗传毒性,并呈一定的剂量-反应关系。
To investigate the effects of cooking soot fine particles on survival rate of fetal mouse alveolar type Ⅱ epithelial cells (AECⅡ) and its genotoxicity. The cells were exposed to different concentrations of PM2.5 for 12 h, 24 h and 48 h, and the cell viability was measured by MTT assay. The tail length, percentage of tail DNA, tail moment and Olive tail moment were measured by comet assay. PAH-DNA adduct content. The results showed that: (1) PM2.5 induced a significant cytotoxicity in dose-response and time-response relationship after exposure to AECⅡ for 12 h, 24 h and 48 h; (2) Compared with low concentration group, , Tail length, tail DNA percentage, tail moment and Olive tail moment of high concentration group increased significantly (P <0.001). Compared with low concentration group, the PHA-DNA adduct of high concentration group was significantly Increase (P <0.05). These results suggest that PM2.5 has obvious genotoxicity to AECⅡ and shows a certain dose-response relationship.
