目的探讨IL-1β对肺腺癌A549细胞CEA表达的影响及IL-1β诱导的CEA对肺癌A549细胞迁移侵袭能力的影响。方法体外培养的A549细胞分别经IL-1β50、100和200 ng/ml刺激24 h,Western-blot和免疫细胞化学方法检验CEA蛋白的表达;采用细胞Transwell实验和侵袭实验检测IL-1β诱导的CEA对肺癌A549细胞迁移和侵袭能力的影响。结果不同浓度的IL-1β(50、100、200 ng/ml)处理A549细胞后,CEA的表达量较空白对照组增高,且呈剂量依耐性(P<0.05);Transwell试验中对照组、100 ng/ml IL-1β组和加兔抗人CEA多抗组穿过Transwell小室膜的细胞数分别为(52.8±5.5)、(117.2±12.2)和(83.6±5.2)个/视野,侵袭试验中对照组、100 ng/ml IL-1β组和加兔抗人CEA多抗组穿过Matrigel胶的细胞数分别为(37.4±6.1)、(92.6±6.7)和(47.8±6.4)个/视野,均为100 ng/ml IL-1β组高于对照组(P<0.05);加兔抗人CEA多抗组低于100 ng/ml IL-1β组(P<0.05)。结论 IL-1β可能通过诱导CEA过表达,促进肺癌A549细胞的迁移和侵袭。
Objective To investigate the effect of IL-1β on CEA expression in lung adenocarcinoma A549 cells and the effect of IL-1β-induced CEA on lung cancer A549 cell migration and invasion. Methods A549 cells cultured in vitro were stimulated with IL-1β50, 100 and 200 ng / ml for 24 h, respectively. The expression of CEA protein was detected by Western-blot and immunocytochemical staining. The transwell assay and invasion assay were used to detect IL-1β-induced CEA On lung cancer A549 cell migration and invasion ability. The levels of IL-1β in 100 ng / ml IL-1β group were lower than those in control group (P <0.05). Conclusion IL-1β may promote the migration and invasion of lung cancer A549 cells by inducing CEA overexpression.
