目的探讨RhoC、PRL-3及FMNL2在结直肠癌(CRC)中的表达及其相互作用关系。方法利用免疫组化、Western blot检测在不同结直肠癌细胞中RhoC、PRL-3蛋白含量变化;用Realtime-RTPCR法分析RhoC、PRL-3mRNA表达情况。结果免疫组化显示FMNL2基因沉默后,RhoC的平均光密度值(AOD)明显下降,而PRL-3的AOD值未见明显差异。随着结直肠癌细胞中FMNL2蛋白水平下调,RhoC蛋白水平表达明显下降,RhoC mRNA的表达水平是SW620细胞的0.618倍,差异有统计学意义(P<0.05),而PRL-3蛋白含量及mRNA的表达水平均未见明显差异。结论 FMNL2可能参与RhoC调控转移机制,而FMNL2对PRL-3的表达可能无反馈性调节作用。
Objective To investigate the expression of RhoC, PRL-3 and FMNL2 in colorectal cancer (CRC) and their interaction. Methods Immunohistochemistry and Western blot were used to detect the protein levels of RhoC and PRL-3 in different colorectal cancer cells. Realtime-RTPCR was used to analyze the expression of RhoC and PRL-3 mRNA. Results Immunohistochemistry showed that the average optical density (AOD) of RhoC decreased significantly after FMNL2 gene silencing, but there was no significant difference in the AOD value of PRL-3. With the downregulation of FMNL2 protein in colorectal cancer cells, RhoC protein expression was significantly decreased, RhoC mRNA expression was 0.618 times higher than that of SW620 cells, the difference was statistically significant (P <0.05), while PRL-3 protein content and mRNA No significant differences in the expression levels were found. Conclusion FMNL2 may be involved in the regulation of RhoC metastasis, but FMNL2 may not have feedback regulation of PRL-3 expression.
