目的改进人外周血淋巴细胞染色体制片技术,探寻一种更简单方便的滴片方法,提高染色体分析效率。方法抽取正常人外周血,按照本实验室经典方案制备淋巴细胞悬液,采用4种不同滴片方法进行滴片,观察并分析不同滴片方法制备的染色体标本效果(重复性及稳定性)。结果采用干冰片过火法获得的可分析染色体分裂相数目丰富,形态结构好,染色体分散面积大,差异小,一个分裂相交叉重叠的染色体少,染色体分裂相破裂的频率低。结论通过本实验研究发现了一种简单可行的有助于染色体最佳扩散的制片技术,大大提高了染色体分析效率。
Objective To improve the technology of chromosomal preparation of human peripheral blood lymphocytes and explore a simple and convenient method of slides to improve the efficiency of chromosome analysis. Methods Peripheral blood samples were collected from normal subjects. Lymphocyte suspensions were prepared according to the classical protocol of our laboratory. Four different methods of slides were used for slides. The effects (repeatability and stability) of chromosomal specimens prepared by different slides methods were observed and analyzed. Results The results showed that the number of chromosomes that can be analyzed by over-fire method is rich, the morphological structure is good, the area of chromosomal scatter is large and the difference is small. There are few chromosomes with overlapping crossover, and the frequency of chromosome cleavage phase rupture is low. Conclusion Through this experimental study, we found a simple and feasible tablet preparation technique that can contribute to the optimal diffusion of chromosomal DNA, and greatly improved the efficiency of chromosome analysis.
