目的 :观察鞘氨醇对人胰腺癌细胞株 JF30 5的增殖作用 ;观察鞘氨醇对人胰腺细胞株 JF30 5内Ca2 + 浓度的影响 ,探讨鞘氨醇的细胞信使作用。方法 :细胞培养后 ,分别加入不同浓度的鞘氨醇 ,应用四甲基偶氮唑盐 MTT比色法来观察细胞增殖程度 ;应用 Fura- 2 / AM荧光比色测定 [Ca2 + ]i 结果 :鞘氨醇促进人胰腺癌细胞株JF30 5的生长 ,促进 JF30 5内 Ca2 +的生成 ,3min内达峰值 ,并在 0 .5 μ m ol/ L~ 2 .0 μ mol/ L 时呈剂量依赖性。结论 :鞘氨醇能促进人胰腺癌细胞株 JF30 5增殖 ;鞘氨醇的信使作用是通过刺激 Ca2 +转运的和 Ca2 +的释放
OBJECTIVE: To observe the effect of sphingosine on the proliferation of human pancreatic cancer cell line JF30 5. To observe the effect of sphingosine on the concentration of Ca2 + in human pancreatic cell line JF305, and to investigate the effect of sphingosine on messenger cell. Methods: After cultured with different concentrations of sphingosine, MTT colorimetry was used to observe the proliferation of cells. Fura-2 / AM fluorescence colorimetry was used to measure the effect of [Ca2 +] i. Sphingosine promoted the growth of human pancreatic cancer cell line JF305 and promoted the formation of intracellular Ca2 + in JF305, reached the peak value within 3 minutes and dose-dependently at the dose of 0.5 μ mol / L ~ 2.0 μ mol / L Sex. Conclusion: Sphingosine can promote the proliferation of human pancreatic cancer cell line JF305. The messenger effect of sphingosine is through the stimulation of Ca2 + transport and Ca2 + release
