糖尿病大鼠肾脏及血清炎症因子的表达

目的探讨血管内皮生长因子(VEGF)、细胞间黏附分子1(ICAM-1)、TNF-α、IL-6和IL-1β在糖尿病大鼠肾脏及血清中的表达差异。方法采用链脲佐菌素诱导27只SD大鼠成功建立糖尿病模型(模型组),分别于成模后2周、2个月采用ELISA法检测肾脏组织及血清中VEGF、TNF-α、ICAM-1、IL-1β和IL-6的含量。另取10只SD大鼠作为对照组,常规饲养2个月取血清及肾脏组织行上述炎症因子的检测。结果模型组成模后2个月,大鼠血清VEGF、TNF-α、ICAM-1、IL-1β和IL-6表达均高于对照组(P<0.05或P<0.01)。模型组成模后2周和2个月,大鼠肾脏中VEGF、TNF-α、ICAM-1、IL-1β和IL-6表达均高于对照组(P<0.05)。模型组成模后2个月,大鼠肾脏中VEGF、IL-1β和IL-6表达高于成模后2周(P<0.05),而TNF-α和ICAM-1表达差异无统计学意义(P>0.05)。结论糖尿病大鼠肾脏及血清中炎症因子VEGF、TNF-α、ICAM-1、IL-1β和IL-6表达均明显增加,且肾脏中VEGF、IL-1β和IL-6表达随病程延长而增加。 Objective To investigate the expression of vascular endothelial growth factor (VEGF), intercellular adhesion molecule 1 (ICAM-1), TNF-α, IL-6 and IL-1β in kidney and serum of diabetic rats. Methods Twenty-seven SD rats induced by streptozotocin (STZ) were successfully established diabetic model (model group). The levels of VEGF, TNF-α, ICAM- 1, IL-1β and IL-6 levels. Another 10 SD rats were taken as the control group, and the serum and kidney tissues were routinely cultured for 2 months to detect the above inflammatory factors. Results The expression of VEGF, TNF-α, ICAM-1, IL-1β and IL-6 in serum of model rats were both higher than those of the control group 2 months after model establishment (P <0.05 or P <0.01). The expression of VEGF, TNF-α, ICAM-1, IL-1β and IL-6 in the rat kidney were both higher than those in the control group at 2 weeks and 2 months after model establishment (P <0.05). The expression of VEGF, IL-1β and IL-6 in kidney of model rats were higher than that of 2 weeks after model establishment (P <0.05), while the expression of TNF-α and ICAM-1 had no statistical significance P> 0.05). Conclusion The expressions of VEGF, TNF-α, ICAM-1, IL-1β and IL-6 in kidneys and serum of diabetic rats were significantly increased, and the expressions of VEGF, IL-1β and IL- .
