[目的]探讨复方苦参注射液(MI)联合小剂量5-氟尿嘧啶(5-Fu)对人胃癌SGC-7901细胞中血管内皮生长因子(VEGF)、趋化因子受体(CXCR4)mRNA及蛋白表达的影响。[方法]采用溴化噻唑蓝四氮唑法观察MI联合小剂量5-Fu对人胃癌SGC-7901细胞增殖的影响;采用逆转录聚合酶链反应法、免疫荧光法、酶联免疫吸附法分别检测MI联合小剂量5-Fu对人胃癌SGC-7901细胞中VEGF、CXCR4 mRNA及蛋白表达水平。[结果]单独应用40μl/ml MI或小剂量(20μg/ml)5-Fu作用于SGC-7901细胞后,与对照组比较,两者均能抑制细胞增殖;20μg/ml5-Fu对VEGF、CXCR4 mRNA水平无明显影响(P>0.05),40μl/ml MI减少VEGF、CXCR4 mRNA及蛋白表达水平(P<0.05);两者联用后能明显降低SGC-7901细胞VEGF、CXCR4 mRNA及蛋白表达水平(P<0.01)。[结论]MI联合小剂量5-Fu具有明显的抑制肿瘤血管生成因子的作用,这可能是MI抗肿瘤血管生成的机制之一。

[Method] The effects of MI combined with low-dose 5-Fu on the proliferation of human gastric cancer cell line SGC-7901 were observed by thiazolyl tetrazolium bromide method. The effects of MI combined with 5-Fu on the proliferation of human gastric cancer cell line SGC-7901 were observed by reverse transcription polymerase chain reaction, immunofluorescence and enzyme-linked immunosorbent assay To detect the expression of VEGF and CXCR4 mRNA and protein in human gastric cancer SGC-7901 cells treated with low dose 5-Fu combined with MI. [Results] After treated with 40μl / ml MI or low dose (20μg / ml) 5-Fu alone, the proliferation of SGC-7901 cells was inhibited by 20μM / ml 5-Fu and VEGF, CXCR4 (P> 0.05) .40μM / ml MI decreased the expression of VEGF and CXCR4 mRNA and protein (P <0.05). The combination of both could significantly reduce the expression of VEGF and CXCR4 mRNA and protein in SGC-7901 cells (P <0.01). [Conclusion] MI combined with low-dose 5-Fu has obvious inhibitory effect on tumor angiogenesis factor, which may be one of the mechanisms of anti-tumor angiogenesis in MI.
